Resazurin Solution:

Composition per 100.0mL:

Resazurin ..........................................................25mg

Preparation of Resazurin Solution: Add resa

zurin to distilled/deionized water and bring volume

to 100.0mL. Mix thoroughly.

Trace Elements Solution:

Composition per 100.0mL:

FeCl3·6H2O.........................................................2.7g

MgSO4 ................................................................1.2g

NaMoO4·2H2O .................................................0.24g

MnSO4·7H2O....................................................0.17g

CaCl2·2H2O ......................................................0.15g

ZnSO4·7H2O..................................................29.0mg

Preparation of Trace Elements Solution: Add

components to distilled/deionized water and bring

volume to 100.0mL. Mix thoroughly.

Sugar Solution:

Composition per 100.0mL:

Xylose.................................................................1.0g

Preparation of Resazurin Solution: Add xy

lose to distilled/deionized water and bring volume to

100.0mL. Mix thoroughly. Filter sterilize.

Preparation of Medium: Add components, ex

cept L-cysteinium chloride, biotin, p-aminobenzoic

acid, and sugar solution, to distilled/deionized water

and bring volume to 995.0mL. Mix thoroughly. Ad

just pH to 7.0. Gently heat but do not boil. Cook for

5-10 min so that color first turns red and then turns

yellow. Cool on ice. Add L-cysteinium chloride, bi

otin, and p-aminobenzoic acid. Mix thoroughly. Au

toclave for 15 min at 15 psi pressure–121°C.

Aseptically add 5.0mL of sterile sugar solution. Mix

thoroughly.

Use: For the cultivation of Clostridium species.
