Phenol Red Carbohydrate Broth

Composition per liter:
Pancreatic digest of casein................................10.0g
NaCl....................................................................5.0g
Beef extract.........................................................1.0g
Carbohydrate solution................................. 200.0mL
Phenol Red solution........................................ 7.2mL
pH 7.4 ± 0.2 at 25°C
Source: This medium is available as a premixed
powder from BD Diagnostics.
Phenol Red Solution:
Composition per 10.0mL:
Phenol Red......................................................0.025g
Preparation of Phenol Red Solution: Add Phe
nol Red to distilled/deionized water and bring vol
ume to 10.0mL. Mix thoroughly.
Carbohydrate Solution:
Composition per 200.0mL:
Carbohydrate..............................................5.0–10.0g
Preparation of Carbohydrate Solution: Add
carbohydrate (5.0g dulcitol, 10.0g lactose, or 10.0g
sucrose) to distilled/deionized water and bring vol
ume to 200.0mL. Mix thoroughly. Filter sterilize.
Preparation of Medium: Add components, ex
cept carbohydrate solution, to distilled/deionized wa
ter and bring volume to 800.0mL. Mix thoroughly.
Adjust pH to 7.4 if necessary. Distribute into tubes
containing an inverted Durham tube. Fill each tube
with 8.0mL of medium. Autoclave for 15 min at 13 psi
pressure–118°C. Cool to 45°–50°C. Aseptically add
2.0mL of sterile carbohydrate solution to each tube.
Use: For the determination of fermentation reactions
in the differentiation of microorganisms, such as Sal
monella spp. Fermentation is determined by the pro
duction of acid—broth turns yellow—and formation
of gas—bubble trapped in Durham tube.
