Composition per liter:

Na2HPO4...........................................................5.64g
NaCl....................................................................5.0g
Pancreatic digest of casein..................................1.5g
Peptic digest of animal tissue .............................1.5g
KH2PO4...........................................................0.225g
KCN solution................................................ 0.15mL
pH 7.6 ± 0.2 at 25°C
Source: This medium is available as a premixed
powder from BD Diagnostic Systems.
KCN Solution:
Composition per 100.0mL:
KCN....................................................................0.5g
Preparation of KCN Solution: Add KCN to
100.0mL of cold distilled/deionized water. Mix thor
oughly and cap. Do not mouth pipette.
Caution: Cyanide is toxic.
Preparation of Medium: Add components, except
KCN solution, to distilled/deionized water and bring
volume to 1.0L. Mix thoroughly. Autoclave for 15 min
at 15 psi pressure–121°C. Cool to room temperature.
Prior to use, add 0.15mL of KCN solution. Mix thor
oughly. Aseptically distribute into sterile tubes.
Use: For the differentiation of Gram-negative
enteric bacteria on the basis of their ability to grow in
the presence of cyanide.
