Composition per liter:

Peptone from casein .........................................20.0g

Agar ..................................................................15.0g

NaCl....................................................................5.0g

Lactose ...............................................................5.0g

K2HPO4...............................................................4.0g

Bile salt mixture..................................................1.5g

KH2PO4...............................................................1.5g

Tryptophan..........................................................1.0g

4-Methylumbelliferyl-ß-D-glucuronide............0.07g

pH 7.0 ± 0.2 at 25°C

Source: This medium is available from Merck.

Preparation of Medium: Add components to

distilled/deionized water and bring volume to 1.0L.

Mix thoroughly. Autoclave for 15 min at 15 psi pres

sure–121°C. Cool to 45°–50°C. Pour into sterile Petri

dishes. The prepared broth is clear and yellowish

brown.

Use: For the detection of E. coli in meats. The medi

um complies with the German-DIN-Norm 10110 for

the examination of meat, with the regulations accord

ing to § 35 LMBG (06.00/36) for the examination of

food and with ISO Standard 6391 (1996) for the enu

meration of E. coli in meat and meat products. The

bile salt mixture of this medium largely inhibits the

accompanying flora not usually found in the intes

tines. Using fluorescence under UV light and a posi

tive indole reaction, E. coli colonies can be identified

among the grown colonies.
