Composition per liter:

Heart muscle ...................................................454.0g
Beef extract.......................................................10.0g
Peptone .............................................................10.0g
NaCl....................................................................5.0g
Glucose ...............................................................2.0g
pH 7.2 ± 0.2 at 25°C
Source: This medium is available as a premixed
powder from Oxoid.
Preparation of Medium: Finely chop beef heart.
Add approximately 1.5g of heart particles to test tubes.
Add remaining components to distilled/deionized wa
ter and bring volume to 1.0L. Mix thoroughly. Distrib
ute into tubes in 10.0mL volumes. Autoclave for 15
min at 15 psi pressure–121°C. Slowly cool tubes to
prevent expulsion of meat particles.
Use: For the cultivation and maintenance of aerobic
and anaerobic microorganisms. For the cultivation of
anaerobes, especially pathogenic Clostridium spp.
