Composition per liter:

Na2HPO4·12H2O ................................................9.0g

NaCl....................................................................5.0g

KH2PO4 ..............................................................1.5g

Meat extract ........................................................1.0g

Yeast extract........................................................1.0g

MgSO4·7H2O......................................................0.2g

MnCl2·4H2O ..................................................20.0mg

CaCl2................................................................1.2mg

Glucose-uric acid solution .......................... 100.0mL

Glucose-Uric Acid Solution:

Composition per 100.0mL:

Glucose ...............................................................5.0g

Uric acid..............................................................0.4g

Preparation of Glucose-Uric Acid Solution:

Add components to distilled/deionized water and

bring volume to 100.0mL. Mix thoroughly. Filter

sterilize.

Preparation of Medium: Add components, ex

cept glucose-uric acid solution, to distilled/deionized

water and bring volume to 900.0mL. Mix thoroughly.

Autoclave for 15 min at 15 psi pressure–121°C. Cool

to 50°–55°C. Aseptically add 100.0mL of sterile glu

cose-uric acid solution. Mix thoroughly. Aseptically

distribute into sterile tubes or flasks.

Use: For the cultivation and maintenance of Bacillus

species.
