Composition per 100.0mL:

L-Tryptophan.......................................................0.5g
NaCl....................................................................0.5g
KH2PO4.............................................................0.25g
pH 7.4 ± 0.2 at 25°C
Preparation of Medium: Add components to
distilled/deionized water and bring volume to
100.0mL. Mix thoroughly. Adjust pH to 7.4. Filter
sterilize. Aseptically distribute in 1.0mL volumes
into sterile screw-capped tubes.
Use: Used to differentiate bacteria based on indole
production. Indole is determined by the addition of
modified Kovacs reagent to cultures that have incu
bated for 18–24 hr. Formation of a red color in the
upper layer indicates indole formation.
