Composition per liter:

Tryptone............................................................20.0g

Agar ..................................................................15.0g

Bile salts No. 3....................................................1.5g

X-glucuronide.................................................0.075g

pH 7.2 ± 0.2 at 25°C

Preparation of Medium: Add components to

distilled/deionized water and bring volume to 1.0L.

Mix thoroughly. Allow to soak for 10 min. Autoclave

for 15 min at 15 psi pressure–121°C. Cool to 45°–

50°C. Pour into sterile Petri dishes.

Use: For the simple enumeration of E. coli without

the need for membranes, or pre-incubation The me

dium has been modified by the addition of a chro

mogenic substrate to detect the β-glucuronidase,

which is highly specific for E. coli. The advantage of

the chromogenic substrate is that it requires no UV

lamp to visualize the reaction, and it is concentrated

within the colony, facilitating easier enumeration in

the presence of other organisms, or when large num

bers are present on the plate.
