Composition per liter:

Na2S2O3 ............................................................30.0g
CaCO3 ...............................................................10.0g
Proteose peptone.................................................5.0g
Bile salts..............................................................1.0g
Iodine solution .............................................. 20.0mL
pH 8.4± 0.2 at 25°C
Source: This medium is available as a premixed
powder from BD Diagnostic Systems.
Iodine Solution:
Composition per 20.0mL:
Iodine ..................................................................6.0g
KI ........................................................................5.0g
Preparation of Iodine Solution: Add iodine and
KI to distilled/deionized water and bring volume to
20.0mL. Mix thoroughly.
Preparation of Medium: Add components, ex
cept iodine solution, to distilled/deionized water and
bring volume to 980.0mL. Mix thoroughly. Gently
heat and bring to boiling. Do not autoclave. Cool to
40°C. Add 20.0mL of iodine solution. Mix thorough
ly. Distribute into tubes in 10.0mL volumes. Use me
dium the same day it is prepared.
Use: For the selective isolation and enrichment of
Salmonella typhi and other salmonellae.
