Lactobacillus leichmannii suspension ............ 1.0mL
pH 6.2 ± 0.1 at 25°C
Preparation of Medium: Add components, ex
cept Lactobacillus leichmannii suspension, to dis
tilled/deionized water and bring volume to 1.0L. Mix
thoroughly. Gently heat and bring to boiling. Auto
clave for 15 min at 15 psi pressure–121°C. Cool to
45°–50°C. Inoculate medium with 1.0mL of Lacto
bacillus leichmannii suspension. Mix thoroughly.
Pour into sterile 150mm Petri dishes in 50.0mL vol
umes. Allow agar surface to dry before using.
Use: For the microbiological assay of vitamin B12 by
the cup plate or disk method using Lactobacillus le
ichmannii as the test microorganism.
